Exhaustion Endpoints

Exhaustion endpoints are predefined behavioral or physiological criteria used to identify when a subject can no longer continue an experimental task safely or reliably. Researchers establish these criteria by monitoring changes such as reduced task performance, loss of coordinated movement, or failure to meet a standardized task demand, then ending the procedure when the endpoint is reached. In behavioral research, exhaustion endpoints help distinguish fatigue-related performance limits from motivation, learning, or disease effects while supporting consistent data collection and humane study design. Clear criteria also improve reproducibility by ensuring that researchers apply comparable stopping rules across subjects and experiments.

In argentometric precipitation titrations, endpoints can be detected visually by the Mohr, Volhard, and Fajans methods. Here, we present a protocol to establish important endpoints and proliferative markers of small intestinal injury and compensatory hyperproliferation using a model of chemotherapy-induced mucositis. We demonstrate the detection of proliferating cells using a cell cycle specific marker and using small intestinal weight, crypt depth, and villus height as endpoints.
