Home Cage Activity Monitoring

Home cage activity monitoring is a behavioral method for measuring spontaneous movement and activity patterns while animals remain in their familiar housing environment. Automated video tracking or infrared beam-break sensors record locomotion, rest, exploration, and activity across light-dark cycles, often continuously and without handling that could alter behavior. In behavioral research, these measurements reveal circadian rhythms, habituation, social or motor changes, and responses to genetic, pharmacological, or environmental manipulation. Because monitoring supports repeated, longitudinal assessment under relatively natural conditions, it can improve detection of subtle phenotypes and provide important context for studies of neurological disease, sleep, stress, and animal welfare.

2015

This report describes a method for measuring Drosophila larval activity using the TriKinetics Drosophila Activity Monitor. The device employs infrared beams to detect movements of up to 16 individual animals. Data can be analyzed to represent motion parameters including rates and the positions of the animals within the assay chambers.

2023

This video describes the Drosophila activity monitor (DAM) system used to track locomotor activity. Researchers use activity data collected from the DAM to study circadian rhythms in fruit flies. The featured protocol clip shows how to load flies in the device and record activity data for circadian experiments.

Isothermal Microcalorimetry to Monitor Metabolic Activity in Bacteria

Take serial dilutions of a bacterial culture and add aliquots into calorimetric vials containing growth medium.Load the sealed vials into the multi-well plate of an isothermal microcalorimeter to measure metabolic heat production. The device is held at a constant temperature to ensure the detected heat originates solely from bacterial metabolism. As bacteria metabolize nutrients and proliferate, they release heat that sensors detect in real time.In parallel, incubate identical vials in an...

Flash Photolysis of Caged Compounds in the Cilia of Olfactory Sensory Neurons

Photolysis of caged compounds allows the production of rapid and localized increases in the concentration of various physiologically active compounds. Here, we show how to obtain patch-clamp recordings combined with photolysis of caged cAMP or caged Ca for the study of olfactory transduction in dissociated mouse olfactory sensory neurons.
