Agricultural Pathogens

Agricultural pathogens are bacteria, fungi, viruses, oomycetes, and other infectious agents that cause disease in crops and livestock, reducing yield and food quality. They initiate infection by attaching to host tissues, entering cells or vascular systems, and using enzymes, toxins, or secreted effector proteins to obtain nutrients and alter host defenses; hosts counter with receptor-mediated immune responses and antimicrobial compounds. Biochemical analysis of these interactions helps identify virulence factors, metabolic pathways, and biomarkers for early detection. This knowledge supports disease-resistant crop breeding, targeted diagnostics, safer pesticide development, and sustainable biological control strategies.

Glass Wool Filters for Concentrating Waterborne Viruses and Agricultural Zoonotic Pathogens

Glass wool filters have been used to concentrate waterborne viruses by a number of research groups around the world. Here we show a simple approach for constructing glass wool filters and demonstrate the filters are also effective in concentrating waterborne viral, bacterial and protozoan pathogens.

Colorimetric Paper-based Detection of Escherichia coli, Salmonella spp., and Listeria monocytogenes from Large Volumes of Agricultural Water

A protocol involving integrated concentration, enrichment, and end-point colorimetric detection of foodborne pathogens in large volumes of agricultural water is presented here. Water is filtered through Modified Moore Swabs (MMS), enriched with selective or non-selective media, and detection is performed using paper-based analytical devices (µPAD) imbedded with bacterial-indicative colorimetric substrates.

Single Cell Measurements of Vacuolar Rupture Caused by Intracellular Pathogens

We describe a method for tracking the endomembrane rupture elicited by the intracellular bacteria Shigella flexneri and Mycobacterium tuberculosis upon host cell invasion. Our assay makes use of CCF4, a host cytoplasmic FRET probe in live or fixed cells. This reporter is degraded by an enzyme activity present on the bacterial surface.
