Detergent Soluble Fraction

The detergent-soluble fraction is the portion of a biological sample whose molecules become dispersed in an aqueous solution after treatment with a detergent, making it useful for separating soluble and membrane-associated components. Detergents disrupt lipid bilayers and weaken hydrophobic interactions, while detergent micelles surround hydrophobic regions and maintain membrane proteins in suspension; centrifugation can then separate this fraction from insoluble material. In biochemistry, analyzing the detergent-soluble fraction supports protein extraction, membrane-protein characterization, subcellular fractionation, and comparisons of protein abundance or activity between soluble and insoluble pools. Its composition depends on detergent type, concentration, temperature, and extraction conditions.
