Lipolysis Measurement

Lipolysis measurement is the quantitative assessment of triglyceride breakdown into glycerol and free fatty acids, providing a direct way to evaluate lipid metabolism in biochemical systems. During lipolysis, lipases hydrolyze ester bonds in triacylglycerols, and researchers measure the resulting glycerol or fatty acid release using biochemical assays under controlled conditions. These measurements help characterize enzyme activity, compare hormonal or nutritional effects on fat mobilization, and investigate metabolic regulation in cells, tissues, and purified preparations. In biochemistry, reliable lipolysis assays support studies of energy balance, obesity, metabolic disease, drug action, and lipid-based biotechnology.

This protocol describes the method of determining basal and forskolin-stimulated lipolysis in inguinal fat pads obtained from normal chow diet (NCD) or high fat diet (HFD) ± capsaicin fed wild type mice. As an index for lipolysis, glycerol release was measured from inguinal adipose fat pads.

Optimized sampling protocols and the development of new wipe materials can be facilitated by standardized measurements of collection efficiency from wipe-sampling. Our approach for sampling trace explosives uses an automated device to control speed, force, and distance during wipe-sampling followed by extraction of collected explosives.
