Nickel Complex Isolation

Nickel complex isolation is the separation and recovery of a nickel-containing coordination compound from a reaction mixture, allowing its composition and chemical behavior to be examined. The process relies on coordination chemistry, in which ligand donor atoms bind nickel and produce a complex with distinctive solubility, stability, and physical properties that support selective precipitation, crystallization, extraction, or purification. In biochemistry, isolated nickel complexes help researchers investigate metal–ligand interactions relevant to metalloenzymes, biomimetic catalysts, and protein-metal binding. Characterizing these compounds can clarify coordination structure, reactivity, and the role of nickel in biological and catalytic processes.

(2017).This video demonstrates the isolation of protein–viral DNA complexes from cell nuclei using click chemistry and magnetic bead purification. The protocol involves labeling viral genomes with alkyne-modified nucleotides, biotinylation via a click reaction, and binding to streptavidin-coated magnetic beads. Following washing and heat elution, the purified...

Source: Laboratory of Dr. Neal Abrams — SUNY College of Environmental Science and Forestry Transition metals are found everywhere from vitamin supplements to electroplating baths. Transition metals also make up the pigments in many paints and compose all minerals. Typically, transition metals are found in the cationic form since they readily oxidize, or lose electrons, and are surrounded by electron donors called ligands. These ligands do not form ionic or covalent bonds with the metal center,...

Multi-component protein complexes play crucial roles during cellular function and development. Here we describe a method used to isolate native protein complexes from Drosophila embryos after in vivo crosslinking followed by purification of the crosslinked complexes for subsequent structure-function analysis.
