Outer Leaflet Lipids

Outer leaflet lipids are the lipid molecules concentrated on the extracellular-facing side of a cell’s plasma membrane, where they help define membrane structure, surface chemistry, and interactions with the environment. Their asymmetric distribution arises during membrane biogenesis and is maintained by selective lipid-translocating enzymes, including flippases, floppases, and scramblases, which move lipids between bilayer leaflets under regulated conditions. Phosphatidylcholine, sphingomyelin, glycolipids, and cholesterol commonly contribute to this surface layer. In biochemistry, studying outer leaflet lipids clarifies membrane organization, cell recognition, signaling, trafficking, apoptosis, and interactions with drugs, toxins, and pathogens.

Protocol for Relative Hydrodynamic Assessment of Tri-leaflet Polymer Valves

There has been renewed interest in developing polymer valves. Here, the objectives are to demonstrate the feasibility of modifying a commercial pulse duplicator to accommodate tri-leaflet geometries and to define a protocol to present polymer valve hydrodynamic data in comparison to native and prosthetic valve data collected under near-identical conditions.

β-barrel outer membrane proteins (OMPs) serve many functions within the outer membranes of Gram-negative bacteria, mitochondria, and chloroplasts. Here, we hope to alleviate a known bottleneck in structural studies by presenting protocols for the production of β-barrel OMPs in sufficient quantities for structure determination by X-ray crystallography or NMR spectroscopy.
