Protein Microcrystals

Protein microcrystals are micron-scale, ordered assemblies of protein molecules that enable structural analysis when producing large single crystals is difficult. They form when purified protein solutions become supersaturated, promoting nucleation and controlled crystal growth under conditions such as adjusted precipitant concentration, pH, temperature, and protein concentration. Because their small size reduces the need for extensive crystal growth, protein microcrystals support techniques such as serial femtosecond crystallography and other diffraction methods that determine three-dimensional molecular structures. In biochemistry, these crystals help reveal active sites, ligand interactions, and conformational changes, advancing studies of enzyme function, molecular recognition, and structure-guided drug development.

Preparation and Delivery of Protein Microcrystals in Lipidic Cubic Phase for Serial Femtosecond Crystallography

We describe procedures for the preparation and delivery of membrane protein microcrystals in lipidic cubic phase for serial crystallography at X-ray free-electron lasers and synchrotron sources. These protocols can also be applied for incorporation and delivery of soluble protein microcrystals, leading to substantially reduced sample consumption compared to liquid injection.

The signal-to-noise ratio of data is one of the most important considerations in performing X-ray diffraction measurements from microcrystals. The VMXm beamline provides a low-noise environment and microbeam for such experiments. Here, we describe sample preparation methods for mounting and cooling microcrystals for VMXm and other microfocus macromolecular crystallography beamlines.

A protocol is presented for X-ray crystallography using protein microcrystals. Two examples analyzing in vivo-grown microcrystals after purification or in cellulo are compared.
