Site-specific Labeling

Site-specific labeling is a biochemical strategy for attaching a detectable or functional probe to one defined location on a protein or other biomolecule, enabling precise molecular analysis. It works by introducing or selecting a unique reactive site, such as an engineered amino acid or cysteine residue, and coupling it to a complementary chemical tag under conditions that limit nonspecific reactions; bioorthogonal reactions can support labeling in complex samples. Fluorescent dyes, affinity handles, and other probes labeled this way help researchers track protein localization, interactions, conformational changes, and activity. The approach improves experimental precision in structural biology, imaging, diagnostics, and biomolecular engineering.

Application of the DNA-Specific Stain Methyl Green in the Fluorescent Labeling of Embryos

A method for fluorescent staining of fixed biological material with the specific DNA label methyl green is described. Methyl green is used in a diluted aqueous solution and is very resistant to photobleaching. Its far-red emission allows for deep specimen imaging, making it particularly adequate for whole embryos.

Here, we present protocols for the identification and purification of ovarian cells from antral follicles. We elaborate on methods for processing whole ovaries for the cryopreservation of cortical strips while also harvesting intact antral follicles that are treated enzymatically to liberate multiple follicle resident cell types, including granulosa, theca, endothelial, hematopoietic, and stromal cells.
