Supercomplex Assessment

Supercomplex assessment is the biochemical evaluation of stable assemblies formed by multiple protein complexes, often within energy-converting membranes. In mitochondrial research, it examines how respiratory-chain complexes associate into higher-order structures by separating membrane protein assemblies under native conditions, followed by immunodetection, mass spectrometry, structural analysis, or measurements of enzymatic activity. These approaches help determine supercomplex composition, organization, stability, and functional relationships among electron transport and ATP-producing pathways. The resulting data support studies of oxidative phosphorylation, membrane bioenergetics, and disease-associated changes, while clarifying whether interactions reflect stable functional units or transient associations.

This protocol describes the separation of functional mitochondrial electron transport chain complexes (Cx) I-V and supercomplexes thereof using native electrophoresis to reveal information about their assembly and structure. The native gel can be subjected to immunoblotting, in-gel assays, and purification by electroelution to further characterize individual complexes.
