Mouse Cell Lines

Mouse cell lines are populations of cells derived from mice and maintained in laboratory culture, providing reproducible systems for studying cellular and biochemical processes. Researchers grow these cells in nutrient media under controlled temperature, gas, and sterility conditions, while selected lines may proliferate indefinitely through spontaneous or engineered immortalization. In biochemistry, mouse cell lines enable analysis of protein expression, enzyme activity, signaling pathways, metabolism, and responses to drugs or genetic manipulation. Their consistent growth and experimental accessibility support mechanistic studies and model aspects of mammalian biology, although results must be interpreted alongside differences between cultured cells and intact tissues.

Isolation and Derivation of Mouse Embryonic Germinal Cells

The ability of embryonic germinal cells to differentiate into primordial germinal cells during early development stages is a perfect model to address our hypothesis about cancer and infertility. This protocol shows how to isolate primordial germinal cells from developing gonads in 10.5-11.5 days post coitum mouse embryos.

An optimized protocol for the isolation of stem cells from the mouse salivary gland is described. The method employs enzymatic and mechanical digestion, and permits isolation of salispheres containing cells with characteristics of stem cells.

Here we show our method to isolate mouse epidermal neural crest stem cells (EPI-NCSC). Technique involves micro-dissecting whisker follicles, isolating the bulge and placeing it into tissue culture. EPI-NCSC start to emigrate from bulge explants onto the substratum within 3 - 4 days.

Generating induced pluripotent stem cell (iPSC) lines produces lines of differing developmental potential even when they pass standard tests for pluripotency. Here we describe a protocol to produce mice derived entirely from iPSCs, which defines the iPSC lines as possessing full pluripotency1.
