Calcium Fluoride Windows

Calcium fluoride windows are optical components made from crystalline calcium fluoride that transmit ultraviolet, visible, and infrared radiation, making them useful wherever light must pass into or out of an instrument. Their broad spectral transmission, low optical absorption, and relatively low refractive index help preserve signal quality while limiting interference from the window material. In bioengineering, these windows support spectroscopy, microscopy, and optical sensor systems used to examine biomolecules, cells, and engineered tissues. Their chemical resistance and suitability for specialized optical assemblies also enable reliable measurements in demanding research environments, although careful handling is needed because calcium fluoride is relatively brittle.

The present protocol describes making a large (6 x 3 mm2) cranial window using food wrap, transparent silicone, and cover glass. This cranial window allows in vivo wide-field and two-photon calcium imaging experiments in the same mouse.
