Histology Processing Cassette

A histology processing cassette is a labeled, perforated container that holds tissue specimens during preparation for microscopic examination. Its openings allow fixatives, dehydrating agents, clearing solvents, and molten paraffin to move through the specimen while the cassette preserves tissue identity and supports handling between processing steps. After fixation and processing, the tissue is embedded in paraffin, sectioned into thin slices, and stained for analysis. In bioengineering, histology processing cassettes help standardize evaluation of engineered tissues, biomaterial responses, and tissue samples by maintaining traceability and consistent preparation, enabling reliable assessment of cellular organization, architecture, and tissue integration.

Pouring and Running a Protein Gel by reusing Commercial Cassettes

Our protocol demonstrates how to pour multiple protein gels at a time by recycling Invitrogen Nupage Novex minigel cassettes, and inexpensive materials purchased at a home improvement store. This economical and streamlined method includes a way to store the gels at 4°C for a few weeks. By re-using the plastic gel cassettes from commercially available gels, labs that run frequent protein gels can save significant costs and help the environment.
