Human Epidermal Keratinocytes

Human epidermal keratinocytes are the predominant cells of the epidermis, where they form the skin’s protective barrier and regulate interactions between the body and its environment. In native tissue, basal keratinocytes proliferate, migrate toward the surface, and undergo differentiation, producing keratin and organizing into stratified layers that culminate in the cornified barrier. In bioengineering, these cells are expanded in culture and incorporated into engineered skin models to study barrier formation, wound repair, dermatological disease, and responses to drugs or chemicals. Their controlled growth and differentiation make them important for developing skin substitutes and more predictive testing platforms.

Human Skin Keratinocyte Isolation: A Method to Culture Primary Keratinocytes from Skin Samples

2023

This video describes the isolation of keratinocytes—the most prominent cells of human skin's epidermal layer, by employing a combination of enzymatic digestion and mechanical disruption methods. This method helps in the preferential growth of keratinocytes over other skin cell types using a selective growth medium.
