In Vivo Organoid

In vivo organoids are three-dimensional, tissue-like structures grown or matured within a living organism, where they can model development, disease, or tissue function more realistically than isolated laboratory cultures. They form when organoid-forming cells are implanted or transplanted into a host and respond to local extracellular-matrix cues, growth factors, cell interactions, and vascular signals that support organization and maturation. In bioengineering, in vivo organoids help evaluate tissue regeneration strategies, study how engineered constructs integrate with host tissues, and assess disease mechanisms or therapeutic responses under physiological conditions. Their use can bridge the gap between in vitro models and complex living systems.

Three-Dimensional Culture Systems: Spheroids and Organoids

2026

Three-dimensional (3D) culture systems, including spheroids and organoids, support New Approach Methodology (NAM)-aligned research by providing more physiologically relevant models than conventional two-dimensional cell cultures. Because they more closely resemble the structure and cellular interactions of tissues and organs, these systems can provide physiologically relevant data before advancing to animal studies. Spheroids as Disease and Drug Screening Models Spheroids are generated when one...
