Peg Da Rings

PEGDA rings are ring-shaped hydrogel structures formed from poly(ethylene glycol) diacrylate, a synthetic, water-compatible polymer used to create defined three-dimensional environments in bioengineering. Their acrylate end groups crosslink when exposed to light in the presence of a photoinitiator, converting a liquid precursor into a stable network whose geometry and stiffness can be adjusted through material composition and fabrication conditions. This tunable architecture supports cell encapsulation, tissue-engineering models, biomaterial testing, and the construction of microscale platforms for studying cell behavior. PEGDA rings therefore provide a reproducible format for investigating how engineered physical environments influence biological organization and function.

The following protocol provides techniques for encapsulating pancreatic β-cells in step-growth PEG-peptide hydrogels formed by thiol-ene photo-click reactions. This material platform not only offers a cytocompatible microenvironment for cell encapsulation, but also permits user-controlled rapid recovery of cell structures formed within the hydrogels.
