Perfusable Microchannel

A perfusable microchannel is a microscale fluid conduit engineered to sustain controlled liquid flow, providing a way to deliver nutrients, remove waste, and reproduce transport conditions in biological systems. Pumps or pressure differences drive culture medium through the channel, while its small dimensions support precise control of flow rate, shear stress, and exposure to dissolved factors. In bioengineering, perfusable microchannels can model blood-vessel transport, connect engineered tissues, and form key components of organ-on-a-chip platforms. By improving mass transfer and enabling continuous perfusion, they help researchers study cell responses, tissue function, disease processes, and therapeutic performance under more physiologically relevant conditions.

Procedure for the Development of Multi-depth Circular Cross-sectional Endothelialized Microchannels-on-a-chip

A microchannels-on-a-chip platform was developed by the combination of photolithographic reflowable photoresist technique, soft lithography, and microfluidics. The endothelialized microchannels platform mimics the three-dimensional (3D) geometry of in vivo microvessels, runs under controlled continuous perfusion flow, allows for high-quality and real-time imaging and can be applied for microvascular research.

This video describes an antegrade perfusion technique of an isolated mouse heart during which a perfusate is injected through the left ventricle, and eventually, it perfuses the entire myocardium. In this video, we perform Langendorff retrograde perfusion of an isolated mouse heart. During the procedure, the cardiac activity of the heart is maintained by perfusing it retrogradely with a perfusion solution via the coronary arteries using a cannula inserted into the ascending aorta. The model is used to study the cardiac pathophysiology of the heart.
