Thymidine Blocking

Thymidine blocking is a cell-cycle synchronization method that temporarily arrests proliferating cells near the G1/S transition, allowing researchers to study cell-cycle-dependent events with greater temporal control. High extracellular thymidine disrupts the balance of deoxyribonucleotide triphosphates, limiting DNA replication; removing thymidine restores nucleotide availability and permits cells to re-enter S phase, while repeated treatment and release can improve synchrony. In bioengineering, this approach supports controlled analysis of DNA synthesis, gene expression, cell division, and responses to biomaterials or engineered microenvironments. It can also improve the timing of experiments involving cell-cycle-dependent production or tissue development.

We have derived a strategy to detect sequential incorporation of thymidine analogues (CldU and IdU) into tissues of adult mice to quantify two successive rounds of cell division. This strategy is useful to detect cell turnover of long-lived tissues, oncogenic transformation, or transit-amplifying cells.

An imaging-based method is described that can be used to identify S-phase and analyze cell cycle dynamics in the C. elegans hermaphrodite germline using the thymidine analog EdU. This method requires no transgenes and is compatible with immunofluorescent staining.
