Tissue Decellularization

Tissue decellularization is a bioengineering process that removes cells and cellular components from donor tissue while preserving the extracellular matrix, creating a scaffold for research and regenerative medicine. Chemical agents such as detergents and enzymes, together with physical treatments, disrupt cell membranes and nucleic acids; controlled processing aims to reduce immunogenic material without damaging the matrix’s three-dimensional structure and biochemical cues. Decellularized tissues can support cell attachment, migration, and differentiation after reseeding with recipient or cultured cells. Applications include engineered organs, tissue repair, disease modeling, and the development of biomaterials that more closely replicate native tissue architecture.

Perfusion decellularization is a novel technique to produce whole liver scaffolds that retains the organ's extracellular matrix composition and microarchitecture. Herein, the method of preparing whole organ scaffolds using perfusion decellularization and subsequent repopulation with hepatocytes is described. Functional and transplantable liver grafts can be generated using this technique.

Here we present, and contrast two protocols used to decellularize plant tissues: a detergent-based approach and a detergent-free approach. Both methods leave behind the extracellular matrix of the plant tissues used, which can then be utilized as scaffolds for tissue engineering applications.
