8-ohdg Staining

8-OHdG staining is a biological technique used to detect 8-hydroxy-2′-deoxyguanosine, a molecular marker of oxidative DNA damage in cells and tissues. During oxidative stress, reactive oxygen species oxidize guanine bases in DNA, generating 8-OHdG; antibodies that recognize this modified nucleoside then reveal its location through immunohistochemical or immunofluorescent labeling. Researchers use staining intensity and distribution to assess oxidative damage in experimental models, cultured cells, and tissue samples. The method supports studies of cellular stress, disease mechanisms, environmental exposures, and the effects of antioxidant or protective treatments, while providing spatial information that complements biochemical measurements.

External electric field induces a voltage on the membrane of a cell, termed the induced membrane voltage (ΔΦ). By using the potentiometric dye di-8-ANEPPS, it is possible to measure the ΔΦ noninvasively. This video shows the protocol for measuring ΔΦ using di-8-ANEPPS.

The method described here allows for the visualization of IL-8 promoter-dependent inflammation activation in the lungs of mice through non-invasive bioluminescence imaging (BLI). The same animal can be subjected to BLI multiple times for up to two months from the time of delivery of the luciferase reporter construct.
