Chip Functionalization

Chip functionalization is the deliberate modification of a chip’s surface or internal structures with chemical groups, polymers, or biomolecules to control interactions with biological samples. It matters because surface properties such as wettability, charge, and molecular recognition strongly influence capture, transport, and signal generation. Functionalization typically uses adsorption or covalent coupling to immobilize probes such as antibodies, enzymes, nucleic acids, or aptamers while preserving their activity under defined assay conditions. In biological techniques, these engineered interfaces support biosensors, cell isolation, drug screening, and lab-on-a-chip systems, improving selectivity and analytical performance for diagnostics and research.

Lensless Fluorescent Microscopy on a Chip

A lensless on-chip fluorescent microscopy platform is demonstrated that can image fluorescent objects over an ultra-wide field-of-view of e.g., >0.6-8 cm2 with <4μm resolution using a compressive sampling based decoding algorithm. Such a compact and wide-field fluorescent on-chip imaging modality could be valuable for high-throughput cytometry, rare-cell research and microarray-analysis.

Chip-Based Digital PCR to Detect Rare Transcript Variants Using a Nanofluidic Chip

This video demonstrates chip-based digital PCR — a variation of the digital PCR technique that is useful in detecting rare transcript variants. The PCR reaction is partitioned into the chambers of a nanofluidic chip, each of which acts as an independent reaction. The detection of fluorescence signals from the chambers with amplified targets confirms the presence of rare transcript variants in the sample.

Microfluidic flow chambers etched by photolithography and fabricated from PDMS are applied to probe functional outcomes associated with EC dysfunction and inflammation. In a representative experiment, the ability of differential shear stress to modulate monocytic cell adhesion to cytokine activated EC monolayers is demonstrated.
