Histidine Tagged Ligand

A histidine-tagged ligand is a ligand engineered with a short sequence of histidine residues, enabling its selective capture, purification, or immobilization in biological experiments. The histidine side chains coordinate metal ions such as nickel or cobalt that are held by chelating groups in affinity resins or biosensor surfaces; excess imidazole or changes in binding conditions can then release the ligand. This strategy supports controlled studies of receptor-ligand interactions, protein assembly, and molecular recognition while simplifying ligand handling. Histidine tagging is also useful for preparing reproducible reagents and analyzing binding specificity in biochemical and cell-based assays.

We report and demonstrate an optimized nitrocellulose binding assay that can be used to quantify autophosphorylation of purified bacterial histidine kinases. Our method has several advantages over traditional SDS-PAGE based techniques, providing a valuable alternative for characterizing these important proteins.
