Macrophage Cell Culture

Macrophage cell culture is the in vitro maintenance and study of macrophages, immune cells that detect threats, engulf particles, and regulate inflammation. In controlled laboratory conditions, macrophages are grown in nutrient media with regulated temperature, pH, and gas exchange; cultures may use primary cells or macrophage-like cells generated from precursor populations. Researchers use these cultures to examine phagocytosis, inflammatory signaling, host-pathogen interactions, and responses to drugs or biomaterials. As a biological technique, macrophage cell culture provides a practical model for investigating innate immunity while reducing the complexity of whole-organism experiments.

A protocol is presented for cell culture of macrophage colony-stimulating factor (M-CSF) differentiated human monocyte-derived macrophages. The protocol utilizes cryopreservation of monocytes coupled with their bulk differentiation into macrophages. Then harvested macrophages can then be seeded into culture wells at required cell densities for carrying out experiments.

A Simple and Efficient Method to Isolate Macrophages from Mixed Primary Cultures of Adult Liver Cells

A novel method to obtain macrophages from primary culture of rat liver cells is described. This method utilizes the proliferation of macrophages in the culture, followed by shaking of culture flasks and purification by selective attachment to plastic dishes. This technique efficiently provides liver macrophages without complex equipment and skills.

Differentiation and Polarization of Monocyte-Derived Cells into Macrophage-Like Cells

In this video, we demonstrate a procedure to induce the differentiation and polarization of peripheral blood-derived monocytes into M1 or M2 macrophages. Incubation with granulocyte-macrophage colony-stimulating factor, lipopolysaccharide, and interferon‐gamma yields pro-inflammatory M1 macrophages, while incubation with macrophage colony-stimulating factor and interleukin-4 induces anti-inflammatory M2 macrophage production.
