Neuroprotective Compound Screening

Neuroprotective compound screening is the systematic evaluation of chemical or biological agents for their ability to preserve neuronal structure and function during disease- or injury-related stress. In biological techniques, researchers expose cultured neurons or other neural models to conditions such as oxidative stress, excitotoxicity, or inflammation, then measure outcomes including cell viability, apoptosis, neurite integrity, and cellular signaling. Screening workflows may progress from rapid in vitro assays to more complex model systems that assess dose response, efficacy, and toxicity. These methods help identify candidate therapeutics for neurodegenerative disorders, stroke, and traumatic neural injury while clarifying mechanisms of neuronal damage and protection.

Evaluating the Neuroprotective Effects of a Test Compound Against Alpha-Synuclein Toxicity in Rat Embryonic Neurons

2025

Start with a primary midbrain culture from rat embryos, containing dopaminergic neurons that overexpress mutated alpha-synuclein.Replace the media in the treatment wells with fresh media containing the test compound, and that in the control wells with plain media. Then incubate. In the control, alpha-synuclein forms toxic aggregates in dopaminergic neurons, causing neurite retraction and leading to neuronal death. In the treatment, the test compound interacts with alpha-synuclein, prevents its...

(2018)This video demonstrates the procedure to assess the neuroprotective effect of a test compound in a middle cerebral artery occlusion mouse model. The compound enters the brain, prevents neuronal death, and reduces infarct volume, confirming its neuroprotective effect compared to an untreated mouse.

2010

We seek to define the neural immune signaling responsible for cold-preconditioning as means to identify novel targets for therapeutics development to protect brain before injury onset. We present strategies for such work that require biological systems, experimental manipulations plus technical capacities that are highly reproducible and sensitive.

A Fluorescence-Based Assay Using Engineered Lymphocytes for Screening Immunomodulatory Compounds

2025

This video demonstrates the screening of immunomodulatory compounds via fluorescent-based assay. Transgenic mouse-derived T lymphocytes with green fluorescent protein expression cassettes are treated with these compounds. The enhanced and reduced GFP signals enable the identification of the compounds' stimulatory and inhibitory effects, respectively.
