Peg Conjugation

PEG conjugation is the covalent attachment of polyethylene glycol (PEG) chains to a molecule, often a protein, peptide, drug, or nanoparticle, to alter its physicochemical and biological behavior. In a typical reaction, chemically activated PEG reacts with nucleophilic groups such as lysine amino groups or cysteine thiols, forming a stable linkage under controlled conditions; PEG can increase hydrophilicity and sterically shield the conjugated molecule. In biological techniques, this modification is used to improve solubility, stability, circulation time, and resistance to enzymatic degradation, supporting protein therapeutics, drug delivery systems, and other biomedical applications.

Here, we present a procedure to fluorescently functionalize the disulfides on Qβ VLP with dibromomaleimide. We describe Qβ expression and purification, the synthesis of dibromomaleimide-functionalized molecules, and the conjugation reaction between dibromomaleimide and Qβ. The resulting yellow fluorescent conjugated particle can be used as a fluorescence probe inside cells.
