3d Intestinal Model

A 3D intestinal model is a laboratory system that reproduces key features of the intestine in three dimensions, providing a more physiologically relevant platform than conventional flat cell cultures. These models commonly use intestinal stem cells or engineered tissues that organize within an extracellular matrix into structures resembling crypts, villi, and a luminal epithelial barrier, sometimes with supporting cell types or controlled fluid flow. Researchers apply 3D intestinal models to study epithelial development, nutrient absorption, host-microbe interactions, intestinal disease, and drug efficacy or toxicity. By capturing tissue architecture and cell-cell interactions, they can improve disease modeling and reduce reliance on animal studies.

Here we describe constructing a basic three-dimensional (3D) intestinal cell line model system and a paraffin embedding protocol for light microscopic evaluation of fixed intestinal equivalents. Staining of selected proteins permits the analysis of multiple visual parameters from a single experiment for potential use in preclinical drug screening studies.

In this video, we demonstrate a technique to isolate whole crypts from a mouse’s small intestine. The isolated whole crypts can be used to propagate long-lived and self-renewing 3D organoids in vitro.

We describe simple methods to study the regulation of intestinal serotonin transporter (SERT) function and expression using an in vitro cell culture model of Caco-2 cells grown in 3D and an ex vivo model of mouse intestines. These methods are applicable to the study of other epithelial transporters.
