Agar Invasion Visualization

Agar invasion visualization is a biological imaging approach that reveals how cells or microorganisms penetrate and spread through a semisolid agar matrix. In this assay, a specimen is introduced onto or within agar, where growth, motility, and matrix penetration produce visible patterns that can be recorded by direct observation, staining, or microscopy. The resulting invasion zones and colony morphologies provide qualitative or quantitative evidence of movement and tissue-like substrate interaction. This method supports studies of microbial behavior, cell migration, virulence, and environmental responses, offering a relatively simple way to compare invasion under different genetic, chemical, or physical conditions.

Assay for Adhesion and Agar Invasion in S. cerevisiae

We describe a qualitative assay for yeast adhesion and agar invasion as a measure of invasive and pseudohyphal differentiation. This simple assay can be used to assess the invasive phenotype of various mutants as well as the effects environmental cues and signaling pathways on yeast differentiation.

Visualization of the Charcoal Agar Resazurin Assay for Semi-quantitative, Medium-throughput Enumeration of Mycobacteria

The charcoal agar resazurin assay (CARA) is a semi-quantitative, medium-throughput method to assess activity of test agents against mycobacteria that are replicating, non-replicating, or both. The CARA permits rapid evaluation of time- and concentration-dependent activity and identifies parameters to pursue by colony forming unit (CFU) assays.
