Agarose Plug Preparation

Agarose plug preparation is a laboratory technique that embeds biological samples, commonly intact cells or genomic DNA, in a solid agarose matrix for handling and analysis. Molten agarose is mixed with the sample and cast in a mold; as it cools, the agarose solidifies around the material, while subsequent in-plug lysis and washing can reduce mechanical shearing of large DNA molecules. These plugs support applications such as pulsed-field gel electrophoresis, restriction analysis, and genome characterization, enabling researchers to examine high-molecular-weight DNA and other specimens with improved structural integrity.
