Antibody Penetration

Antibody penetration is the movement of antibody molecules through biological barriers and into tissues, tumors, or cells, where it determines how effectively they can reach and bind their targets. Penetration depends on molecular size, charge, affinity, tissue structure, vascular permeability, and transport processes such as diffusion, convection, and receptor-mediated uptake; dense extracellular matrices and limited blood flow can restrict distribution. Studying these factors helps explain uneven antibody localization, supports the design of immunostaining and imaging protocols, and guides the development of antibody-based diagnostics and therapeutics with improved tissue access and target engagement.

The goal of this protocol is to alter the penetrance of lethal skeletal mutant phenotypes in zebrafish by selective breeding. Lethal mutants cannot be grown to adulthood and bred themselves, therefore this protocol describes a method for tracking and selecting penetrance through multiple generations by progeny testing.

Inducing Penetrating Traumatic Brain Injury in Adult Drosophila Flies

This video demonstrates the induction of penetrating traumatic brain injury in the drosophila brain. Newly emerged flies are selected, anesthetized, and positioned under a fluorescence-equipped stereo microscope. The GFP-expressing mushroom body is identified, and a Minutien pin is inserted to cause injury. The flies are allowed to recover for further studies.
