Apomorphine Rotation Test

The apomorphine rotation test is a behavioral assay used to measure asymmetric dopamine signaling, especially in animal models of Parkinson’s disease. After a unilateral lesion of the nigrostriatal pathway, commonly induced with 6-hydroxydopamine, apomorphine stimulates dopamine receptors and produces rotational behavior because the damaged and intact sides respond differently; the direction and rate of rotation reflect this imbalance. Researchers use rotation counts to estimate lesion severity, evaluate dopaminergic dysfunction, and assess the effects of neuroprotective or restorative treatments. Although the test provides a practical measure of motor asymmetry, results depend on lesion extent, drug dose, and experimental conditions.
