Arteriolar Tone

Arteriolar tone is the baseline degree of contraction in arteriolar smooth muscle, a key determinant of vascular resistance, blood pressure, and tissue blood flow. It arises from a balance among sympathetic nerve activity, circulating hormones, endothelial signals, and local metabolic conditions that regulate smooth-muscle contraction and relaxation. Increasing tone narrows the vessel and raises resistance, whereas relaxing the muscle promotes vasodilation and greater perfusion. In biology, understanding arteriolar tone helps explain cardiovascular homeostasis, regional blood-flow regulation, autoregulation, and disorders such as hypertension, in which abnormal vascular constriction can impair organ function.

We have developed a device (Twister) to study the regulation of tonic muscle activity during active postural maintenance. Twister measures torsional resistance and muscular responses in standing subjects during twisting of the body axis. The device can be flexibly configured to study various aspects of tonic control across the neck, trunk, and/or hips.

Intensive preparation of intact mouse cerebral endothelial "tubes" from cerebral parenchymal arterioles is illustrated for studying cerebral blood flow regulation. Further, we demonstrate the experimental strengths of this endothelial study model for fluorescence imaging and electrophysiology measurement of key cellular signaling pathways, including changes in intracellular [Ca2+] and membrane potential.
