Automated Video Tracking

Automated video tracking is a computer-based technique that detects and follows moving organisms, cells, or other biological objects across sequential video frames, converting visual movement into quantitative data. Image-analysis algorithms identify target features, distinguish them from the background, and link detections over time to reconstruct trajectories, speed, distance, or behavioral events. In biology, the method supports objective measurement of locomotion, animal behavior, cell migration, and interactions while reducing manual scoring. By enabling high-throughput, reproducible analysis of complex movements, automated video tracking helps researchers compare phenotypes, evaluate experimental treatments, and investigate biological mechanisms.

The loss of honey bee colonies presents a challenge to crop pollination services. Current pollinator protection practices warrant an alternative approach to minimize the contact of honey bees to harmful pesticides using repellent chemistries. Here, we provide detailed methods for a visual tracking protocol to screen deterrents for bees.

Locomotor activity (LMA) is a simple and easily performed measurement of behavior in mice. Coupling of video tracking software (VTS) and LMA allows for the improvement of specificity and sensitivity, especially when compared with the manual, line crossing method of LMA analysis. Additionally VTS allows long-term tracking of mouse LMA.

We describe a protocol of brightfield intravital microscopy for measuring dynamic neutrophil-endothelial cell interactions during neutrophil recruitment in response to the source of a neutrophil chemoattractant in vivo. Neutrophil intraluminal crawling, transendothelial migration and chemotaxis in mouse cremaster muscle tissue are visualized with time-lapsed video photography and tracked with ImageJ.
