Bacterial Culture

Bacterial culture is the laboratory practice of growing bacteria in a controlled medium, making otherwise microscopic populations available for observation and study. A sample is introduced into sterile liquid or solid growth medium, then incubated under conditions such as suitable temperature, oxygen availability, and nutrient supply; bacterial cell division produces turbidity in broth or visible colonies on agar. Researchers use bacterial culture to isolate organisms, examine colony and growth characteristics, maintain biological stocks, and obtain material for microscopy or downstream experiments. In biology, this method supports investigations of microbial physiology, ecology, genetics, and interactions with their environments.

Source: Lin, P., et al. Synthesizing Sodium Tungstate and Sodium Molybdate Microcapsules via Bacterial Mineral Excretion. J. Vis. Exp. (2018).This video demonstrates the preparation of a monoclonal bacterial culture for mineral production through serial dilution and colony isolation on an agar plate. By shaking the plate with glass beads, individual cells are spatially separated to form genetically uniform colonies, which are then transferred to liquid media for growth. The culture with the...

(2017)This video demonstrates the preparation of a fungal culture on treated cellophane membranes to support colony formation and radial growth. It then shows the cultivation and preparation of soil bacterial cells for interaction assays. Finally, it illustrates how bacterial cells adhere to the fungal colony...

Exp.(2015)This video demonstrates the preparation of a bacterial culture for bacteriophage production. It covers the steps of inoculating a soil bacterium into a baffled flask, promoting uniform growth through aeration and shaking, and monitoring cell density using optical density measurements. Once the culture reaches the optimal growth phase,...

A Technique to Establish a Bacterial Infection Model in an Ex Vivo Organ Culture

2025

This video demonstrates a method to establish an infection model in placental tissues to study infection propagation. An ex vivo placental organ culture is infected with Listeria monocytogenes, a facultative intracellular parasite. The bacteria invade extravillous trophoblasts (EVTs) to enter the placental tissue and then propagate to other cells via cell-to-cell spread.
