Bacterial Keratitis Model

A bacterial keratitis model is an experimental system used to study bacterial infection and inflammation of the cornea, the transparent outer surface of the eye. It reproduces key features of disease by exposing corneal tissue or ocular cells to pathogenic bacteria, allowing researchers to examine microbial attachment, invasion, host immune responses, and tissue damage under controlled conditions. These models support investigation of virulence factors, antimicrobial treatments, and mechanisms of corneal injury. By connecting bacterial behavior with epithelial disruption and inflammatory outcomes, they help evaluate therapies and improve understanding of a vision-threatening ocular infection.

Porcine Ex vivo Cornea Model of Bacterial Keratitis: A Technique to Establish Bacterial Infection in Corneal Epithelial Cells

2025

This video describes a protocol to set up an ex vivo model for bacterial keratitis using a corneoscleral button dissected from the porcine eye. Inoculated bacteria traverse the corneal epithelial incisions to invade the underlying stroma and establish an infection within the corneal tissues that mimic the in vivo infection.

Recurrent Herpetic Stromal Keratitis in Mice, a Model for Studying Human HSK

Most studies of herpetic corneal disease use a primary infection model. However, primary infection with HSV-1 does not typically lead to human disease. Here we describe a recurrent model of herpetic corneal disease, which more closely mimics human disease.

Establishing a Porcine Ex Vivo Cornea Model for Studying Drug Treatments against Bacterial Keratitis

This article describes a step-by-step protocol to set up an ex vivo porcine model of bacterial keratitis. Pseudomonas aeruginosa is used as a prototypic organism. This innovative model mimics in vivo infection as bacterial proliferation is dependent on the ability of the bacterium to damage corneal tissue.

Bacterial Infection Quantitation Assay: A Method to Quantify Intraocular Bacterial Load in Mouse Model of Bacterial Endophthalmitis

2025

This video describes the technique of quantifying bacterial infection from eye homogenates obtained from a murine model of bacterial endophthalmitis caused by Bacillus cereus. This technique helps study the host immune response and its dependency on the host and ocular bacterial gene expression in murine models.

Bacterial Endophthalmitis Mouse Model: A Method for Generating Bacterial Endophthalmitis via Intravitreal Injection of Bacterial Suspension into Eye of Mouse

2025

This video describes a method for generating a mouse model that mimics bacterial endophthalmitis – an intraocular infection resulting from a bacterial invasion into the eye’s vitreous cavity – via intravitreal injection of a bacterial suspension into the eye. This mouse model helps study the disease mechanism and evaluate therapeutics for it.
