Beta-sheet Aggregation

Beta-sheet aggregation is the self-association of protein or peptide molecules into assemblies enriched in repeating beta-sheet structures, a process important in molecular biology because it can alter protein function and produce stable cellular deposits. Misfolded or partially unfolded chains expose aggregation-prone sequences, which align through backbone hydrogen bonding and form nuclei that elongate into fibrils or larger aggregates. In biology, beta-sheet aggregation is central to studies of amyloid diseases, including Alzheimer’s and Parkinson’s disease, where aggregates can disrupt cellular processes. Understanding its kinetics, structural transitions, and environmental triggers also supports biomarker development, therapeutic screening, and the design of protein-based materials.

We describe a method of measuring binding energy, expressible as tissue surface tension, between cells within 3D tissue-like aggregates. Differences in tissue surface tension have been demonstrated to correlate with invasiveness of lung, muscle, and brain tumors, and are fundamental determinants of establishing spatial relationships between different cell types.
