Bisulphite Conversion

Bisulphite conversion is a molecular technique used to distinguish methylated from unmethylated cytosines in DNA, making it central to DNA methylation and epigenetic analysis. During treatment, bisulphite chemically converts unmethylated cytosine residues into uracil, while 5-methylcytosine is largely protected from conversion; after amplification and sequencing, these differences appear as thymine or cytosine signals. Researchers use the method in bisulphite sequencing and methylation-specific assays to map methylation patterns across genes and genomes, helping investigate gene regulation, development, disease-associated epigenetic changes, and other biological processes.
