Chlorophyll A B Ratio

The chlorophyll a/b ratio is a quantitative measure of the relative amounts of chlorophyll a and chlorophyll b in photosynthetic organisms, providing insight into their light-harvesting systems and adaptation to available light. Chlorophyll a functions as the primary reaction-center pigment, while chlorophyll b expands the range of absorbed wavelengths and transfers excitation energy to chlorophyll a within antenna complexes. Because shade-grown leaves typically invest more in light-harvesting pigments, their chlorophyll a/b ratio often differs from that of sun-exposed leaves. Measuring this ratio supports studies of photosynthetic performance, plant stress, acclimation, algal ecology, and the responses of vegetation to changing environmental conditions.

This manuscript describes a simple and high-throughput method for assembling water-soluble proteins with hydrophobic pigments that is based on water-in-oil emulsions. We demonstrate the effectiveness of the method in the assembly of native chlorophylls with four variants of recombinant water-soluble-chlorophyll binding proteins (WSCPs) of Brassica plants expressed in E. coli.
