Chlorophyll A Measurement

Chlorophyll a measurement is the quantitative assessment of chlorophyll a, the primary photosynthetic pigment found in algae, cyanobacteria, and plants, and a widely used indicator of photosynthetic biomass and aquatic productivity. In common spectrophotometric methods, pigments are extracted from cells or tissues with an organic solvent, absorbance is measured at chlorophyll a-specific wavelengths, and established equations convert those readings into concentration; fluorometric methods instead detect chlorophyll fluorescence. In biology, these measurements support studies of photosynthesis, phytoplankton abundance, water quality, ecosystem productivity, and environmental change, while helping researchers compare biological responses across samples and conditions.

Water in Oil Emulsions: A New System for Assembling Water-soluble Chlorophyll-binding Proteins with Hydrophobic Pigments

This manuscript describes a simple and high-throughput method for assembling water-soluble proteins with hydrophobic pigments that is based on water-in-oil emulsions. We demonstrate the effectiveness of the method in the assembly of native chlorophylls with four variants of recombinant water-soluble-chlorophyll binding proteins (WSCPs) of Brassica plants expressed in E. coli.

We describe a new technical approach to study photosynthetic responses in higher plants involving simultaneous measurements of chlorophyll a fluorescence and leaf reflectance using a PAM and a spectral radiometer for the detection of signals from the same leaf area in Arabidopsis.

The protocol introduces a high-throughput method for measuring the relaxation of non-photochemical quenching by pulse amplitude modulated chlorophyll fluorometry. The method is applied to field-grown Glycine max and can be adapted to other species to screen for genetic diversity or breeding populations.
