Collagenase Elastase

Collagenase elastase refers to proteolytic enzymes that break down collagen and elastin, the structural proteins that give connective tissues strength and elasticity. Collagenase cleaves collagen fibers within the extracellular matrix, while elastase hydrolyzes elastin and other protein substrates, loosening the tissue framework and enabling cell release. In biology, these enzymes are used to dissociate tissues for primary cell isolation, culture, and experimental analysis, including studies of tissue organization and cellular behavior. Controlled enzyme concentration, exposure time, and temperature help balance efficient tissue digestion with preservation of cell viability and function.

This video shows how to induce abdominal aortic aneurysms (AAA) in mice via transient intraluminal infusion of porcine pancreatic elastase into the infrarenal segment of the abdominal aorta. The model has the ability to add broad insight into the pathobiology of AAA due to the emergence of numerous transgenic and gene knockout mice.

Developing an Experimental Aneurysm Model in a Rabbit Using an Elastase-Treated Vessel Pouch

Source: Wanderer, S., et al. Arterial Pouch Microsurgical Bifurcation Aneurysm Model in the Rabbit. J. Vis. Exp. (2020)This video demonstrates the creation of an experimental aneurysm model in a rabbit using an elastase-treated vessel pouch. After excising a segment of the right CCA, the pouch is treated with elastase, then sutured to the left CCA to restore blood flow. The resulting stress on the weakened pouch walls induces aneurysm formation.

2014

Preclinical models of intracerebral hemorrhage are utilized to mimic certain aspects of clinical disease. Thus, mechanisms of injury and potential therapeutic strategies may be explored. In this protocol, two models of intracerebral hemorrhage are described, intrastriatal (basal ganglia) injections of autologous blood or collagenase.

Isolation of Human Hepatocytes by a Two-step Collagenase Perfusion Procedure

A modified two-step collagenase perfusion procedure for isolation of human hepatocytes is described. This method can also be applied to other mammalian livers. The isolated hepatocytes are available in high yield and viability, making them a suitable model for scientific research in areas such as liver regeneration, pharmacokinetics and toxicology.
