Compound Microscopy Imaging

Compound microscopy imaging is a light microscopy technique that uses multiple lenses to produce a magnified, resolved image of small specimens, making cellular structures and microorganisms visible in biology. Light passes through or reflects from a prepared sample, while the objective lens forms the primary image and the ocular lens further magnifies it; resolution depends on optical design, illumination, specimen preparation, and focusing. Researchers use compound microscopes to examine cell morphology, tissue organization, microbial growth, and staining patterns. The method provides an accessible foundation for observing biological structure and complements advanced approaches such as fluorescence and electron microscopy.

This article describes a working protocol to image dendritic spines from hippocampal neurons in vitro using Structured Illumination Microscopy (SIM). Super-resolution microscopy using SIM provides image resolution significantly beyond the light diffraction limit in all three spatial dimensions, allowing the imaging of individual dendritic spines with improved detail.
