Cryopreservation Eggs

Egg cryopreservation is a reproductive biology technique that preserves oocytes at ultra-low temperatures for later use, extending the time in which viable eggs can support research or assisted reproduction. In the most widely used approach, vitrification exposes mature oocytes to cryoprotective solutions and rapidly cools them in liquid nitrogen, forming a glass-like state that limits ice-crystal formation and cellular damage. Controlled warming then removes the cryoprotectants and prepares the eggs for fertilization or analysis. This method supports fertility preservation, in vitro fertilization, and studies of oocyte quality, development, and reproductive aging.

This is a high-throughput sperm cryopreservation protocol for zebrafish. Sperm cryopreserved using this protocol has an average of 25% fertility in subsequent vitro fertilization and is stable over many years.

Mouse Sperm Cryopreservation and Recovery using the I·Cryo Kit

Here we demonstrate the newly developed I•Cryo kit for mouse sperm cryopreservation. Two-cell stage embryo development with frozen-thawed sperm was improved consistently in 5 mouse strains with the use of this kit. Over a 1.5 year period, 49 genetically modified mouse lines were archived by sperm cryopreservation with the I•Cryo kit and later successfully recovered by IVF.

Isolation and Cryopreservation of Neonatal Rat Cardiomyocytes

The isolation of neonatal rat cardiomyocytes is a time consuming and unpredictable procedure. This study describes methods for cryopreservation and thawing of neonatal rat cardiomyocytes that allows for more efficient use of cells. The thawed NRCMs can be used for various experiments without the need for performing isolations each time.

Drosophila Egg Collection and Dechorionation: A Method to Remove the Outermost Egg Layer

The Drosophila embryo is surrounded by protective outer membranes. To gain access to the embryo, the chorion–the outermost of the membranes–is often removed. This video describes a commonly used method of dechorionation using sodium hypochlorite. The example protocol demonstrates the procedure while employing sterile techniques necessary for axenic or gnotobiotic animal rearing.
