Cytosolic Calcium Measurement

Cytosolic calcium measurement is the quantitative assessment of free calcium ions in a cell’s cytoplasm, a key approach for studying signaling, secretion, contraction, metabolism, and cell death. Researchers typically use fluorescent chemical indicators or genetically encoded calcium indicators that change their fluorescence after binding Ca2+, allowing changes in intracellular concentration to be monitored with microscopy or plate-based instruments. Measurements can reveal rapid calcium dynamics, including transient spikes, oscillations, and sustained elevations, in response to hormones, neurotransmitters, electrical activity, or cellular stress. In biology, these data help link calcium signals to cell function, disease mechanisms, drug responses, and physiological regulation.

A quick, simple method using a flow cytometer to determine relative changes in cytosolic calcium in adherent epithelial cells brought into suspension was optimized.

This video describes the technique of imaging islet cells in real-time, to study the changes in cytosolic calcium concentration in the minor subpopulations of the pancreatic islet cells. Minor cell subpopulations can be identified using functional responses to marker compounds, such as adrenaline and ghrelin, that induce changes in cytosolic calcium in a specific populations of islet cells. This can provide an insight into the cross talk between the blood glucose and the islet secretory output.

Vacuolar and cytosolic pH can be measured in live yeast (S. cerevisiae) cells using ratiometric fluorescent dyes localized to specific cellular compartments. We describe procedures for measuring vacuolar pH with BCECF-AM, which localizes to the vacuole in yeast, and cytosolic pH with a cytosolic ratiometric pH-sensitive GFP (yeast pHluorin).
