Detached Leaf Bioassay

A detached leaf bioassay is an ex vivo biology method that evaluates plant responses under controlled conditions after a leaf is removed from the parent plant. The excised leaf is placed on a moist support and exposed to a pathogen, chemical, hormone, or other treatment, while researchers measure outcomes such as lesion development, chlorosis, tissue damage, or changes in growth. By isolating leaf tissues from whole-plant influences, this assay enables rapid comparisons of host susceptibility, disease progression, phytotoxicity, and treatment effects. It is useful for screening microbial interactions and candidate compounds before more extensive greenhouse or field experiments.

Unraveling how physiology and morphology are linked allows a deeper understanding of mechanistic functioning of plant leaves. We present both a procedure to derive parameters of stomatal regulation from stomatal conductance measurements and correlations with traditional functional leaf traits.

A Simple Bioassay for the Evaluation of Vascular Endothelial Growth Factors

We describe a simple cell-based bioassay for detecting, quantifying and monitoring the activity of members of the vascular endothelial growth factor family of ligands. The assay uses chimeric receptors expressed in a factor-dependent cell line to provide a semi-quantitative or quantitative assessment of receptor binding and cross-linking by the ligand.
