Embryo Bisection

Embryo bisection is a micromanipulation technique that divides an early embryo into two portions, potentially producing genetically identical offspring or embryo halves for biological research. Typically performed at the morula or blastocyst stage, the procedure uses a microsurgical blade to separate the embryo through its cellular mass while preserving developmental potential and, when possible, the capacity of each portion to form a viable embryo. In developmental biology, embryo bisection helps investigate early cell organization, lineage formation, and developmental competence. It also supports embryo multiplication in livestock breeding and assisted reproduction, although outcomes depend on embryo quality, stage, and successful post-bisection culture.

Plastic sections maintain true tissue morphology in thin sections of tissue that can be immunostained with fluorescent secondary antibodies, making this method more useful than paraffin-embedded or frozen sections for many types of tissue. The method for staining, plastic embedding, and sectioning is demonstrated in this video.

Here, we present a protocol to significantly reduce the mitochondrial DNA copy numbers in a bovine oocyte (P < 0.0001). This method utilizes centrifugation and bisection to substantially reduce oocyte mitochondria and may allow for an increased chance of development in the reconstructed interspecies somatic cell nuclear transfer embryos.

2023

This video describes how to perform microinjections that deliver reagents into live Drosophila embryos. Researchers can use this method to introduce exogenous compounds like nucleic acids and proteins during the early stages of embryo development. The featured protocol demonstrates how to set up and perform the procedure for injection of any soluble reagent–here, a fluorescent protein for live imaging experiments.
