Endogenous Virus Isolation

Endogenous virus isolation is the recovery and characterization of virus particles or viral genomes that originate from viral sequences embedded in a host organism’s genetic material, a process that helps distinguish inherited viral elements from active infections. Researchers typically examine host cells or tissues, apply conditions that can activate integrated proviral sequences, and detect resulting viral RNA, proteins, particles, or replication in permissive cultures, often confirming identity through molecular sequencing. This approach reveals whether endogenous elements remain replication competent, clarifies their effects on host biology and immunity, and supports studies of viral evolution, genome integration, zoonotic emergence, and potential contamination of biological research materials.

Begin with a tube containing anesthetized adult Drosophila of the same sex and species.Add a buffer and homogenize it to release endogenous viral populations and bacteria.Centrifuge the homogenate, then pass the supernatant through a membrane by centrifugation to remove bacteria and debris.Transfer the filtrate to a multi-well plate containing engineered Drosophila cell cultures that are permissive to RNA viruses and incubate. The viruses enter the host cell via receptor-mediated...

Numerous novel virus-like sequences have been found in mosquitoes due to the extensive use of sequencing technologies. We provide an effective procedure for isolating and amplifying viruses using vertebrate and mosquito cell lines, which might serve as the basis for future studies on mosquito-associated viruses, including mosquito-borne and mosquito-specific viruses.
