Fetal Liver Isolation

Fetal liver isolation is a biological technique used to remove and study liver tissue from a developing fetus under controlled laboratory conditions. The procedure typically involves carefully separating the fetal liver from surrounding tissues, dissociating it into individual cells or small tissue fragments, and maintaining the material in an appropriate culture or analysis system. Because fetal liver contains developing hepatocytes, hematopoietic progenitors, and other stromal cell populations, isolation enables researchers to examine liver development, blood-cell formation, cellular differentiation, and tissue-specific gene expression. These preparations also support studies of developmental disorders, regeneration, toxicology, and the cellular mechanisms that shape organ maturation.

Isolation of Leukocytes from the Murine Tissues at the Maternal-Fetal Interface

Described herein is a protocol to isolate and analyze the infiltrating leukocytes of tissues at the maternal-fetal interface (uterus, decidua, and placenta) of mice. This protocol maintains the integrity of most cell surface markers and yields enough viable cells for downstream applications including flow cytometry analysis.

The porcine model of liver normothermic machine perfusion (NMP), described here, can be successfully used to study NMP as a preservation strategy, a tool for viability assessment, and a platform for organ repair. It holds a high translational value, however it is technically challenging and labor-intensive.

Isolation of Rat Portal Fibroblasts by In situ Liver Perfusion

A technique for isolating portal fibroblasts from rat liver is described. Livers are perfused and digested in situ with collagenase, followed by ex vivo digestion of the liver slurry and size selection of cells. This method provides a pure population of portal fibroblasts without the need for passage in culture.
