Ffpe Biopsy Tissue

FFPE biopsy tissue is biological tissue preserved by formalin fixation and paraffin embedding, a widely used method for maintaining biopsy specimens for histological and molecular analysis. Formalin cross-links cellular proteins to stabilize tissue architecture, while dehydration and infiltration with molten paraffin create a firm block that can be sectioned into thin slices for staining and microscopic examination. FFPE samples support routine diagnosis, immunohistochemistry, and extraction of DNA or RNA for selected molecular assays. Because archived blocks can retain clinically valuable information for years, they also enable retrospective studies of disease features, biomarkers, and treatment responses.

We describe a method to establish human enteroids from small intestinal crypts and colonoids from colon crypts collected from both surgical tissue and biopsies. In this methodological article, we present the culture modalities that are essential for the successful growth and maintenance of human enteroids and colonoids.
