Fitc Dapi Filters

FITC DAPI filters are optical components used in fluorescence microscopes to selectively illuminate and detect samples labeled with fluorescein isothiocyanate (FITC) or 4′,6-diamidino-2-phenylindole (DAPI), enabling researchers to distinguish molecular signals from background light. Each filter set uses an excitation bandpass filter, a dichroic mirror, and an emission filter: FITC typically emits green fluorescence near 520 nanometers, while DAPI emits blue fluorescence near 460 nanometers after ultraviolet excitation. In biology, these sets support immunofluorescence, nuclear staining, cell counting, and localization of proteins or nucleic acids. Correct spectral matching improves contrast, reduces bleed-through, and helps researchers quantify spatial patterns in cells and tissues.

This video demonstrates a method to study permeation through an in vitro blood-brain barrier (BBB) model using astrocytes and endothelial cells on a membrane insert, with free FITC and FITC-loaded ferritin. FITC-ferritin bound to receptors on endothelial cells and crossed the membrane barrier to the lower chamber while free FITC showed restricted permeation and remained in the upper chamber.

A method to establish an in vitro model of blood-brain barrier based on a co-culture of rat brain microvascular endothelial cells and astrocytes is described and validated. This system proved to be a valid tool to study the effect of nanoformulation on the trans-barrier permeation of fluorescent molecules.
