Fluorescent Antibodies

Fluorescent antibodies are antibody molecules linked to fluorophores, light-emitting labels that enable researchers to detect and locate specific antigens in biological samples. When the antibody binds its target through antigen-antibody recognition, the fluorophore absorbs light at an excitation wavelength and emits light at a longer wavelength, producing a measurable signal. In biology, these reagents support immunofluorescence microscopy, flow cytometry, and fluorescence-based assays for identifying proteins, characterizing cells, and tracking cellular structures or processes. Directly labeled antibodies provide a signal from the primary antibody, whereas secondary fluorescent antibodies can amplify detection and offer flexibility across experiments.

The on-bead method for labeling antibodies with small molecules enables labeling of a small amount of antibodies directly from cell media. This method is compatible with amine and thiol chemistry, and can handle multiple samples in parallel, manually or using automated platforms.

In Vivo Fluorescence Imaging to Localize Antibodies in a Mouse Tumor Xenograft Model

2025

The video demonstrates a fluorescence imaging method for in vivo antibody localization in a mouse tumor xenograft model. It involves the injection of tumor cells in a mouse to generate solid xenografts beneath the skin. Subsequent injection of fluorescent antibodies confirms specific localization through enhanced fluorescence signals within the tumor xenograft.

We described a procedure for the disaggregation of colorectal cancer (CRC) to produce viable single cells, which are then captured on customized antibody microarrays recognizing surface antigens (DotScan CRC microarray). Sub-populations of cells bound to the microarray can be profiled by fluorescence multiplexing using monoclonal antibodies tagged with fluorescent dyes.
