Fluorescent Lipid

A fluorescent lipid is a lipid molecule linked to a fluorophore, a light-sensitive group that enables researchers to visualize lipid behavior in biological systems. When illuminated at an appropriate excitation wavelength, the fluorophore absorbs energy and emits light at a longer wavelength, allowing the labeled lipid to be detected by fluorescence microscopy. Because fluorescent lipids can associate with cellular membranes, they help reveal membrane organization, lipid transport, vesicle trafficking, and interactions between membranes and proteins. These probes support studies of cell structure, signaling, membrane dynamics, and the spatial distribution of lipids in living or fixed biological samples.

A technique to probe the lipid raft partitioning of fluorescent proteins at the plasma membrane of living cells is described. It takes advantage of the disparity in diffusion times of proteins located inside or outside of lipid rafts. Acquisition can be performed dynamically in control conditions or after drug addition.

Here, we describe a protocol to obtain the lipid droplet index (LD index) to study the dynamics of triacylglycerols in cells cultured in high-throughput experiments. The LD index assay is an easy and reliable method that uses BODIPY 493/503. This assay does not need dispendious lipid extraction or microscopy analysis.
